Microbiólogo y patólogo estadounidense. Se licenció de médico en la Universidad de Cornell. Descubrió que la inyección de cultivos inactivados por el calor, de los microorganismos causantes de ciertas enfermedades, podía inmunizar a los animales contra la enfermedad correspondiente. Al descubrir que la fiebre de Texas del ganado es producida por un parásito transmitido por garrapatas (la primera demostración clara del papel de los artrópodos en la diseminación de enfermedades) contribuyó a que la comunidad científica aceptara el rol de los mosquitos en el paludismo y en la fiebre amarilla. Smith fue el primero en diferenciar las bacterias que causan la tuberculosis del ganado y de los humanos, y uno de los primeros en percatarse de la anafilaxis. También mejoró la producción de vacunas en el laboratorio.
